Effects of the Kanamycin & Gentamicin of Hair Cells of Corti¢¥s Organ of Guinea Pigs

Abstract
The aim of this study was to investigate morphologically, using LM & EM, the modes of sensory cell damage of organ of Corti experimentally caused by ototoxc antibiotics.
Healthy 63 young guinea pigs, weighing 200-350g each were used and divided into 3 groups: saline treated, K-M intoxicated and G-M intoxicated. The subjects were again subdivided into subgroups, based on the daily doses of druge received: 15, 50 and 100 §·/§¸ B.W. for K-M; 40, 80 and 160 §·/§¸ B.W for G-M.
Each animal was subcutaneously injected drugs daily for 1, 3 and 6 wks and sacrificed immediately after the last injection.
Our LM & EM approaches for the histopathological study were the surface preparation technique, using 0.2% AgNO_(3) stain and the surface block technique, using, the fixation with mixed solution of 2.5% glutaraldehyde -2% paraformaldehyde & 1% osmium tetroxide.
The results were as followings:
1) Sensory cell degeneration was first observed in 6th wok-15 §·/§¸ B.W & 50 §·/§¸ B.W and also in 3rd wk-100 §·/§¸ B.W experimental groups for the K-M intoxication: in 3rd wk-40 §·/§¸ B.W & in 1st wk-80 §·/§¸ B.W experimental groups for the G-M intoxication.
2) In both K-M & G-M intoxication, sensory cell degeneration, was frequently observed in OHCs of all turns proportionately to their intoxicated dosage and duration. The degeneration was more frequent in the 1st row and severe in the basal turn. Damage in IHCs was also observed, especially in the apical turn.
3) The LM findings revealed phalTangeal scars or collapsed figures as well as complete loss of hair cells as a sign of degeneration in the cells.
4) The EM findings revealed cytoplasmic protrusion into endolymphatic space and the appearance of lysosomes & multivesiculated bodies. Formation of myelin figures in the supranuclear cytoplasm was also revealed together with vesicles & vacuoles in the cytoplasm of the degenerating hair cells.
